heal.abstractΣτην παρούσα βιβλιογραφική εργασία μελετάται φαρμακολογικά και σε θεωρητικό επίπεδο η πιθανή δημιουργία μιας νέας φαρμακοτεχνικής μορφής σταθερού συνδυασμού ιβουπροφαίνης και ομεπραζόλης. Σκοπός της εργασίας είναι να διερευνηθεί η φαρμακολογική συμβατότητα των δύο δραστικών ουσιών, ώστε αυτές να συμπεριληφθούν στην ίδια φαρμακοτεχνική μορφή. Οι μελέτες για την δημιουργία σκευασμάτων σταθερού συνδυασμού δύο ή περισσότερων δραστικών ουσιών παρουσιάζουν συνεχώς αυξανόμενο ενδιαφέρον, λόγω των πολλών πρακτικών, κοινωνικών αλλά και οικονομικών προτερημάτων τους. Έτσι, για τον σκοπό της εργασίας, παρουσιάζονται εκτενώς τα φαρμακοδυναμικά και φαρμακοκινητικά χαρακτηριστικά της ιβουπροφαίνης και της ομεπραζόλης, ώστε να εξαχθεί ασφαλές συμπέρασμα για το αν οι δύο ουσίες μπορούν να συγχορηγηθούν αποτελεσματικά και με τι κόστος. Πιο συγκεκριμένα, παρουσιάζεται αναλυτικά ο μηχανισμός δράσης του κάθε μορίου, οι επιθυμητές και ανεπιθύμητες ενέργειες, οι χρήσεις και οι ενδείξεις, οι αλληλεπιδράσεις με άλλα φάρμακα ή συμπληρώματα διατροφής ή τρόφιμα. Ακόμη, παρατίθενται τα φαρμακοκινητικά τους προφίλ, δηλαδή η έναρξη και η διάρκεια της δράσης τους, η απορρόφηση, η κατανομή, ο μεταβολισμός και η απέκκριση της κάθε μία ουσίας ξεχωριστά. Για την εξαγωγή των συμπερασμάτων, τα προφίλ τους συγκρίνονται και παρουσιάζονται οι ιδιότητες και τα χαρακτηριστικά του πιθανού συνδυασμού τους, καθώς και οι πιθανές αλληλεπιδράσεις αυτού με άλλα φάρμακα, συμπληρώματα διατροφής ή τρόφιμα. Τα ευρήματα της εργασίας ήταν ενθαρρυντικά ως προς τη δημιουργία του πιθανού συνδυασμού, καθώς δεν διαπιστώθηκε, σε θεωρητικό επίπεδο, κάποια ασυμβατότητα μεταξύ των δύο ουσιών. Ενθαρρυντικό είναι, ακόμη, το γεγονός ότι ήδη κυκλοφορεί στην Ευρώπη και την Αμερική σκεύασμα σταθερού συνδυασμού ναπροξένης και ισομεπραζόλης, δραστικές ουσίες που ανήκουν στις ίδιες φαρμακολογικές ομάδες με την ιβουπροφαίνη και την ομεπραζόλη. Καταλήγοντας, για την έγκριση του νέου σκευάσματος είναι απαραίτητη η διεξαγωγή μελετών βιοϊσοδυναμίας (in vitro και in vivo) που θα επιβεβαιώσουν και θα κατοχυρώσουν και κλινικά τα συμπεράσματα της παρούσας μελέτης.el
heal.abstractIn this bibliographic diploma thesis, the possible formulation of a new pharmaceutical form of a fixed dose combination of ibuprofen and omeprazole is being studied pharmacologically and at a theoretical level. The purpose of this research is to investigate the pharmacological compatibility of the two active substances, so that they can be included in the same pharmaceutical form. The studies for the formulation of stable combinations of two or more active substances show an ever-increasing interest, due to their many practical, social and economic advantages. Thus, for the purpose of this project, the pharmacodynamic and pharmacokinetic characteristics of ibuprofen and omeprazole are extensively presented, to draw a safe conclusion on whether the two substances can be co-administered effectively and at what cost. More specifically, the mechanism of action of each molecule, the desired and undesired effects, the uses and indications, the interactions with other drugs or nutritional supplements or food are presented in detail. Furthermore, their pharmacokinetic profiles are listed, i.e., the onset and duration of their action, the absorption, distribution, metabolism, and excretion of each individual substance. To draw the conclusions, their profiles are compared, and the properties and characteristics of their possible combination are presented, as well as its possible interactions with other drugs, nutritional supplements or food. The findings of the research were encouraging regarding the formulation of the possible combination, as no incompatibility between the two substances was found, on a theoretical level. It is also encouraging that in Europe and America there is already a fixed dose combination formulation of naproxen and esomeprazole, active substances that belong to the same pharmacological groups as ibuprofen and omeprazole. In conclusion, for the approval of the new preparation it is necessary to carry out bioequivalence studies (in vitro and in vivo) that will confirm and validate the conclusions of the present study clinically.en
