| heal.abstract | Η παρούσα εργασία πραγματεύεται το θέμα της αλληλούχισης νέας γενιάς στον καρκίνο. Η εργασία ολοκληρώνεται μέσα από οκτώ κεφάλαια. Αναλυτικά το πρώτο κεφάλαιο εστιάζει στον προσδιορισμό της αλληλούχισης του DNA κατά Sanger. Συγκεκριμένα παρουσιάζει την μέθοδο, την αλληλούχιση χρώσης-τερματιστή, την αυτοματοποίηση και προετοιμασία των δειγμάτων, τις προκλήσεις, τον προσδιορισμό αλληλούχισης κατά Sanger με μικρορευστά και τις εφαρμογές τεχνολογιών προσδιορισμού αλληλούχισης με μικρορευστά. Επίσης περιγράφεται ο σχεδιασμός συσκευών, η χημεία προσδιορισμού αλληλουχιών, οι πλατφόρμες και οι συγκρίσεις με άλλες τεχνικές προσδιορισμού αλληλούχισης. Το δεύτερο κεφάλαιο εστιάζει στην NGS, μία τεχνολογία αλληλούχισης επόμενης γενιάς. Το τρίτο κεφάλαιο παρουσιάζει τα κληρονομικά σύνδρομα καρκίνου και το τέταρτο κεφάλαιο αναλύει τη σωματική μετάλλαξη του καρκίνου καταλήγοντας σε συμπερασματικές παρατηρήσεις σχετικά με Gene-Panels για την ανάλυση σωματικών μεταλλάξεων και για τις αναλύσεις της υγρής βιοψίας. Το πέμπτο κεφάλαιο μελετά την φαρμακογενετική και το έκτο εστιάζει σε άλλες εφαρμογές και μελλοντικές κατευθύνσεις. Το έβδομο κεφάλαιο παρουσιάζει τους περιορισμούς και τις επιπλοκές του NGS στα γενετικά διαγνωστικά και ηθικά ζητήματα. Τέλος το όγδοο κεφάλαιο καταλήγει σε συμπερασματικές παρατηρήσεις. | el |
| heal.abstract | This paper addresses the issue of new generation sequencing especially in cancer. It consists of eight chapters. The first chapter focuses on Sanger DNA sequencing. Specifically, it presents the method, the dyeterminator sequencing, the automation and sample preparation, the challenges, the Sanger sequencing with microfluids and the applications of microfluids in sequencing technologies. It describes the pattern, sequencing chemistry, platforms, and comparisons with other sequencing techniques. The second chapter focuses on NGS, a next-generation sequencing technology. The third chapter presents inherited cancer syndromes and the fourth chapter analyzes cancer somatic mutation concluding with remarks about Gene-Panels for the analysis of somatic mutations and the technology of liquid biopsy. The fifth chapter presents pharmacogenetics and the sixth focuses on other applications and future directions. Chapter seven presents the limitations and complications of NGS in genetic diagnostics and ethics. Finally, the eighth chapter concludes with observations. | en |
