| heal.abstract | Η παρούσα εργασία εμβαθύνει στον καρκίνο και ειδικότερα, στον καρκίνο του μαστού. Αρχικά, εξετάζει τις αντινεοπλασματικές θεραπείες, συμβατικές και πιο καινοτόμες, εστιάζοντας στη θεμελιώδη αξία της επαναστόχευσης, ήδη, ευρέως χρησιμοποιούμενων φαρμάκων, σε νέα πεδία εφαρμογής, όπως η αξιοποίηση των αντιψυχωσικών φαρμάκων στην αντινεοπλασματική θεραπεία. Για τον σκοπό αυτό, αναδεικνύονται εκτενώς οι ακολουθούμενες πειραματικές πορείες in vitro αξιολόγησης των αντικαρκινικών ιδιοτήτων αντιψυχωσικών, είτε ως μονοθεραπεία είτε σε σχήμα συγχορήγησης, με συμβατικά αντινεοπλασματικά φάρμακα. Έτσι, με εφαρμογή Κυτταρομετρίας Ροής, Ανοσοαποτύπωσης κατά Western και άλλων μεθόδων, εξετάζεται η επίδραση των αντιψυχωσικών σε κυτταρικά σηματοδοτικά μονοπάτια, σχετιζόμενα με την καρκινογένεση. Τα αποτελέσματα παρουσιάζουν τις αντικαρκινικές ιδιότητες αντιψυχωσικών, όπως η αριπιπραζόλη, η χλωροπρομαζίνη και η πιμοζίδη και τον μείζονα ρόλο τους στη ρύθμιση της έκφρασης αντιαποπτωτικών πρωτεϊνών. Ως απότοκο, τα αντιψυχωσικά παρουσιάζονται ως υποψήφιοι αντινεοπλασματικοί παράγοντες, ωστόσο απαιτείται περαιτέρω έρευνα, τόσο σε in vivo όσο και σε in vitro επίπεδο, για την εδραίωσή τους στην κλινική πράξη. | el |
| heal.abstract | This paper delves into cancer and, in particular, breast cancer. First, it examines antitumor therapies, both conventional and more innovative, focusing on the fundamental value of retargeting already widely used drugs to new fields of application, such as the use of antipsychotic drugs in antineoplastic therapy. For this purpose, the following experimental courses of in vitro evaluation of the antitumor properties of antipsychotics, either as monotherapy or in co-administration with conventional antineoplastic drugs, are highlighted extensively. Thus, by applying Flow Cytometry, Western Blot and other methods, the effect of antipsychotics on cellular signaling pathways related to carcinogenesis is examined. The results indicated the antitumor properties of antipsychotics, including aripiprazole, chlorpromazine and pimozide and their major role in regulating the expression of antiapoptotic proteins. It is of interest to note that antipsychotics could be considered as candidate antineoplastic agents, however thorough research studies, both preclinical and clinical, are needed to establish them in clinical practice. | en |
