Αναστολή της χημικής καρκινογένεσης από τροποποιητές της συσσώρευσης αιμοπεταλίων (Doctoral thesis)

ΤHE AIM OF THIS STUDY WAS TO EXAMINE THE MODIFICATIVE EFFECT OF SUBSTANCES WHICH INTERFERE WITH PLATELET AGGREGATION, IN CARCINOGENESIS INDUCED BY BAP BOTH IN EXPERIMENTAL (WISTAR RATS) AND IN CONTROL ANIMALS. THE PROBABLE ROLE OF CYTOKINES HAS ALSO BEEN INVESTIGATED IN THREE GROUPS OF EXPERIMENTS: 1. EXPERIMENTS FOR INHIBITION OF CARCINOGENESIS INDUCED BY BAP IN WISTAR RATS WITH MODIFICATORS OF PLATELET AGGREGATION. 2. PLATELET AGGREGATION TESTS USING AGONISTS AND INHIBITORS OF AGGREGATION WHICH HAVE BEEN USED AS ANTICARCINOGENS. 3. MEASUREMENTS OF CYTOKINES IN SERUM AND IN SUPERNATANT OF PBL CULTURES IN PATIENTS WITH NEOPLASIAS, USING ANTIOXIDANTS. CONCLUSIONS: 1) ANTIOXIDANTS CAUSE INHIBITION OF CHEMICAL CARCINOGENESIS, INDUCED BY BAP IN EXPERIMENTAL ANIMALS, WHEN THEY ARE GIVEN BEFORE THE ADMINISTRATION OF CARCINOGEN. 2) ANTIOXIDANTS CAUSE INHIBITION OR STIMULATION OF PLATELETS AGGREGATION. 3) THE COMMON ENZYMATIC SYSTEM INVOLVED IN THESE TWO PROCESSES - CARCINOGENESIS AND PLATELET AGGREGATION -SEEMS TO BE THE CYCLOXYGENASE - PROSTAGLANDIN SYNTHETASE WHICH IS PART OF CYTOCHROME P450 ENZYMES. 4) THE EFFECT OF ANTIOXIDANTS IS NOT INDUCED BY CYTOKINES. 5) ANTIOXIDANTS ACT VIA A FREE RADICAL BINDING MECHANISM. 6) ATOXIC NUTRITIONAL PROGRAMMES OF MINIMAL COST CAN BE DESIGNED TO PREVENT THE CARCINOGENESIS AND THROMBOSIS, TWO MAIN CAUSES OF DEATH IN THE WEST.
