Ανίχνευση και συγκριτική μελέτη στελεχών του ιού παπιλλώματος σε λεμφαδένες και νεοπλάσματα αιδοίου κόλπου και τραχήλου μήτρας (Doctoral thesis)

THE PURPOSE OF THIS STUDY IS THE DETECTION OF HUMAN PAPILLOMAVIRUS IN CARCINOMAS OF THE ULERINE CERVIX AND THE POSSIBLE ASSOCIATION BETWEEN HPV INFECTIONAND P53 ACCUMULATION IN CANCEROUS CELL. THE PRESENCE OF HPV AND THE PREVALENCE OF DIFFERENT HPV GENOTYPES WAS DEMONSTRATED BY SOUTHERN BLOT ANALYSIS, PCR ANALYSIS AND IN SITU DNA HYBRIDIZATION. ALSO, THE EXPRESSION OF P53 PROTEIN WAS IMMUNOHISTOCHEMICALLY ANALYSED. OUT OF 81 CASES, HPV DNA WAS DETECTED IN 59,3% OF THE SAMPLES. POSITIVELY FOR HPV WAS DISTRIBUTED IN 0,9% FOR HPV6, 2,8% FOR HPV11, 33,4% FOR HPV16, 16,7% FOR HPV18, 3,7% FOR HPV31 AND 12% FOR HPV33. WE FOUND ALSO AN INEXPECTABLY HIGH PERCENTAGE (37,5) OF HPV MULTIPLE INFECTION IN CIN III AND SQUAMOUS CELLARCINOMA SPECIMENTS. P53 WAS DETECTED IN 67,9% OF THE CASES, BUT LOW-LEVEL P53 ACCUMULATION WAS IDENTIFIED IN HPV16 INFECTED SPECIMENTS. ON THE OTHER HAND, HIGH-LEVEL P53 ACCUMULATION WAS IDENTIFIED IN HPV-MULTIPLE INFECTED COUCEROUS SAMPLES. THERE IS NO SIGNIFICANT ACCOSIATION BETWEEN THE PRESENCE OF HPV AND METASTASIS OR CLINICAL COURCE OF THE DISEASE. FINALLY, HPV IS A CRUCIAL EVENT IN CERVICAL CARCINOGENESIS, BUT ITS ROLE IS STILL UNDER INVESTIGATION AND IS RELATED TO THE FUNCTION OF OTHER MOLECULES. IN GREECE, THERE IS A PREVALENCE OF HPV MULTIPLE INFECTION (37,5%), INCONTRAST TO OTHER EPIDEMIOLOGICAL DATA FROM DIFFERENT COUNTRIES.
